Tryptic Soy Agar (BD 236950) ……………….… 40.0 g
DI Water……………………………………….…. 950.0 mL
Broth Medium
Tryptic Soy Broth (BD 211825) …………………. 30.0 g
DI Water……………………………………...…... 950.0 mL

Autoclave medium at 121ºC. Cool to ~47ºC.

Aseptically add:
Defibrinated Sheep Blood (room temperature)…….... 50.0 mL

Gently mix and dispense into appropriate vessel type.
To make medium from scratch, use the formulation below:
Tryptic Soy Agar/Broth Composition
Tryptone…………………………………….. 15.0 g
Soytone……………………………………… 5.0 g
NaCl…………………………….................... 5.0 g
*Agar……………………………................... 15.0 g
DI Water…………………………………….. 950.0 mL
*Omit agar from broth formulation
pH to 7.3 +/- 0.2.
Note: Tryptone-Pancreatic Digest of Casein or Soytone-Papaic Digest of Soybean Meal
can be used in place of Tryptone and Soytone respectively.
Final Product Description
➢ Agar should be bright red.
➢ For broth, blood will settle to the bottom;
once shaken, broth should be bright red.
➢ Recommended Storage Temperature: 2-8ºC.
