Description

 2-Mercaptoethanol (also known as β-mercaptoethanol, 2-mercaptoethanol, or simply ME) is an organic compound with the chemical formula HOCH₂CH₂SH. It is a colorless liquid with a strong, unpleasant odor. The compound contains a thiol (-SH) group and is commonly used as a reducing agent in biochemical and molecular biology applications. The thiol group reacts with disulfide bonds in proteins, breaking them and resulting in a reduction of protein structure.

Common Uses in a Biology Lab

  • Protein denaturation: 2-Mercaptoethanol is widely used to break disulfide bonds in proteins, aiding in protein denaturation for techniques like SDS-PAGE (sodium dodecyl sulfate-polymerase chain reaction) and protein electrophoresis. This helps to linearize the proteins and separate them based on molecular weight.
  • Cell culture: It can be used in cell culture media to prevent oxidation and stabilize proteins. Its reducing properties help maintain the integrity of thiol groups in cell growth factors, ensuring that they remain active in cell culture environments.
  • Nucleic acid studies: 2-Mercaptoethanol is used in nucleic acid purification protocols to reduce disulfide linkages in nucleases, thus preventing the degradation of RNA and DNA. It is commonly used in RNA extraction kits to protect the RNA from oxidative damage.
  • Elution of proteins from columns: In protein purification, 2-mercaptoethanol is used to elute proteins from affinity columns, as it helps in breaking the disulfide bonds and facilitating protein release.
  • Buffering agent in electrophoresis: It is added to sample buffers to protect proteins from oxidative damage during electrophoresis and to keep them in their reduced form.
